چکیده

The brief publication [1] in Biochimica et Biophysica Acta (BBA) entitled "Enzymic Synthesis of Deoxyribonucleic Acid", submitted May 2, 1956, was the first such report of DNA synthesis, aside from an abstract which appeared in Federation Proceedings in March of that year [2]. In the BBA paper, we (see photograph, p. 55) showed that the incorporation of [a4C]thymidine into an acid-soluble, DNase-sensitive, alkali-stable product depended on partially purified enzyme fractions, ATP and DNA which was converted to an 'active primer'. We expressed the brash hope of assembling 'genetically specific DNA'. Two years later, we submitted two papers to the Journal of Biological Chemistry describing the 2000-fold purification of DNA polymerase and the enzymatic synthesis of the four deoxyribonucleoside triphosphates which the enzyme assembled into long chains. The papers were titled: 'The Enzymatic Synthesis of DNA. I. Preparation of Substrates and Partial Purification of an Enzyme from Escheriehia coli. II. General Properties of the Reaction.' The papers were not accepted because of the term 'DNA' in the title. The consensus among the many referees after a protracted review was that the papers would be acceptable if we used the term "polydeoxyribonucleotide" to identify the synthetic product; the use of 'DNA' could be justified only with a demonstration of genetic activity. To us this seemed unreasonable, because few if any DNA papers in the Journal had been required to fulfill this criterion. Fortunately, with accession of John Edsall to the editorship, the papers were accepted with DNA in the title [3,4].

SPECIFIC AMPLIFICATION OF ASPERGILLUS FUMlGATUS DNA BY POLYMERASE CHAIN REACTION

Invasive aspergillosis (1 is a life-threatening condition in immunocompromised patients. An early diagnosis is of great importance because early treatment may resolve this potentially fatal infection. Recently, the polymerase chain reaction (PCR) has been used successfully in detecting specific DNA of several pathogen. In this study, nested PCR was used to detect DNA specific for A!.pergiflus s...

DETECTION AND RESTRICTION ANALYSIS OF C YTOMEGALOVIRUS DNA PERSISTING IN HUMAN ATHEROSCLEROTIC PLAQUES USING POLYMERASE CHAIN REACTION

The polymerase chain reaction (PCR) as applied to detection of a foreign DNA in clinical specimens could provide a sensitive instrument for rapid detection of viral DNA persisting in tissues of patients suspected of latent infection. Human cytomegalovirus (HCMV) DNA was found in arterial plaques of patients with atherosclerotic lesions using a PCR assay with nested primer oligonucleotides ...

RAPID DETECTION OF MYCOBACTERIUM TUBERCULOSIS IN CLINICAL SPECIMENS BY POLYMERASE CHAIN REACTION

We investigated the use of DNA amplification by polymerase chain reaction (peR) for detection of Mycobacterium tuberculosis in 300 patients who were suspected of having pulmonary tuberculosis and compared the results with culture results which were performed in parallel with PCR. Two-thirds of each sample was processed for smear and culture by standard methods and one-third was prepared fo...

Association between Insulin–Like Growth Factor I Polymorphism and Early Growth Traits in Iranian Zandi Sheep, Found Polymerase Chain Reaction Restriction Fragment Length Polymorphism (PCR-RFLP)

Early growth traits in sheep are economically important. Breeding for these traits has been shown to be problematic using quantitative genetic methods, particularly in native sheep herds. Molecular genetics is useful for sheep breeding. The gene for insulin-like growth factor 1 (IGF1) is one of the important candidate genes known for ovine early growth traits. The aim of this study was to inves...

EVALUATION OF DIRECT MICROSCOPY, CULTURE, AND POLYMERASE CHAIN REACTION FOR THE DIAGNOSIS OF TUBERCULOUS MENINGITIS

In view of the importance of early diagnosis of tuberculous meningitis (TBM), the efficiency of the polymerase chain reaction (PCR), one of the most reliable and sensitive DNA-based assays, was compared with conventional methods (acid-fast microscopy and culture) for the detection of M. tuberculosis in cerebrospinal fluid (CSF) specimens from patients suspected of TBM. Of the 29 CSF specim...

P-244: Analysis of Genomic and Cell Free DNA of A let-7 microRNA Binding Site of KRAS Gene Polymorphisms in Endometriosis

Background: Endometriosis is one of the most common benign gynecological diseases which is characterized by endometriallike tissue growing outside the uterine cavity. Although the pathology of endometriosis remains unknown, the genetic predisposition plays an apparent role. Several genes have been contributed to endometriosis, but it seems KRAS has a crucial role, because its activation results...
